ABSTRACT

Abstract Objective Due to the popularity of 3D technology, surgeons can create specific surgical guides and sterilize them in their institutions. The aim of the present study is to compare the efficacy of the autoclave and ethylene oxide (EO) sterilization methods for objects produced by 3D printing with polylactic acid (PLA) material. Methods Forty cubic-shaped objects were printed with PLA material. Twenty were solid and 20 were hollow (printed with little internal filling). Twenty objects (10 solid and 10 hollow) were sterilized in autoclave, forming Group 1. The others (10 solid and 10 hollow) were sterilized in EO, composing Group 2. After sterilization, they were stored and referred to culture. Hollow objects of both groups were broken during sowing, communicating the dead space with the culture medium. The results obtained were statistically analyzed (Fisher exact test and residue analysis). Results In group 1 (autoclave), there was bacterial growth in 50% of solid objects and in 30% of hollow objects. In group 2 (EO), growth occurred in 20% of hollow objects, with no bacterial growth in solid objects (100% of negative samples). The bacteria isolated in the positive cases was non-coagulase-producing Staphylococcus Gram positive. Conclusions Sterilization by both autoclave and EO was not effective for hollow printed objects. Solid objects sterilized by autoclave did not demonstrate 100% of negative samples and were not safe in the present assay. Complete absence of contamination occurred only with solid objects sterilized by EO, which is the combination recommended by the authors.


Resumo Objetivo Devido à popularidade da tecnologia 3D, cirurgiões podem criar guias cirúrgicos específicos e esterilizá-los nas suas instituições. O objetivo do presente estudo é comparar a eficácia dos métodos de esterilização por autoclave e óxido de etileno (OE) de objetos produzidos pela impressão 3D com material ácido polilático (PLA, na sigla em inglês). Métodos Quarenta objetos em formato cúbico foram impressos com material de PLA. Vinte eram sólidos e 20 eram ocos (impressos com pouco enchimento interno). Vinte objetos (10 sólidos e 10 ocos) foram esterilizados em autoclave, formando o Grupo 1. Os demais (10 sólidos e 10 ocos) foram esterilizados em OE, compondo o Grupo 2. Após a esterilização, os objetos foram armazenados e encaminhados para cultura. Objetos ocos de ambos os grupos foram quebrados durante a semeadura, comunicando o espaço morto com o meio de cultura. Os resultados obtidos foram analisados estatisticamente (teste exato de Fisher e análise de resíduo). Resultados No grupo 1 (autoclave) houve crescimento bacteriano em 50% dos objetos sólidos e em 30% dos objetos ocos. No grupo 2 (OE) o crescimento ocorreu em 20% dos objetos ocos, com ausência de crescimento bacteriano nos objetos sólidos (100% de amostras negativas). A bactéria isolada nos casos positivos foi Staphylococcus Gram positivo não produtor de coagulase. Conclusões A esterilização tanto em autoclave quanto pelo OE não foi eficaz para objetos impressos no formato oco. Objetos sólidos esterilizados em autoclave não demonstraram 100% de amostras negativas, não sendo seguro no presente ensaio. Ausência completa de contaminação ocorreu apenas com objetos sólidos esterilizados pelo OE, sendo a combinação recomendada pelos autores.

ABSTRACT

Organic acids are organic compounds that can be synthesized using biological systems. They often contain one or more low molecular weight acidic groups, such as carboxyl group and sulphonic group. Organic acids are widely used in food, agriculture, medicine, bio-based materials industry and other fields. Yeast has unique advantages of biosafety, strong stress resistance, wide substrate spectrum, convenient genetic transformation, and mature large-scale culture technology. Therefore, it is appealing to produce organic acids by yeast. However, challenges such as low concentration, many by-products and low fermentation efficiency still exist. With the development of yeast metabolic engineering and synthetic biology technology, rapid progress has been made in this field recently. Here we summarize the progress of biosynthesis of 11 organic acids by yeast. These organic acids include bulk carboxylic acids and high-value organic acids that can be produced naturally or heterologously. Finally, future prospects in this field were proposed.

ABSTRACT

Plastics are one of the most important polymers with huge global demand. However, the downsides of this polymer are that it is difficult to degrade, which causes huge pollution. The environmental-friendly bio-degradable plastics therefore could be an alternative and eventually fulfill the ever-growing demand from every aspect of the society. One of the building blocks of bio-degradable plastics is dicarboxylic acids, which have excellent biodegradability and numerous industrial applications. More importantly, dicarboxylic acid can be biologically synthesized. Herein, this review discusses the recent advance on the biosynthesis routes and metabolic engineering strategies of some of the typical dicarboxylic acids, in hope that it will help to provide inspiration to further efforts on the biosynthesis of dicarboxylic acids.

Like other agricultural countries, bulk quantities of watermelon peels (WMP) are disposed-off in environment as waste in Pakistan and appropriate management of this waste is the need of hour to save environment from pollution. The work emphasizes the role of ethanologenic yeasts to utilize significant sugars present in WMP for low-cost bioethanol fermentation. Dilute hydrochloric acid hydrolysis of WMP was carried out on optimized conditions employing RSM (response surface methodology) following central composite design (CCD). This experimental design is based on optimization of ethanologenesis involving some key independent parameters such as WMP hydrolysate and synthetic media ratio (X1), incubation temperature (X2) and incubation temperature (X3) for maximal ethanol yield exploiting standard (Saccharomyces cerevisiae K7) as well as experimental (Metchnikowia cibodasensisY34) yeasts. The results revealed that maximal ethanol yields obtained from S. cerevisiae K7 was 0.36±0.02 g/g of reducing sugars whereas M. cibodasensisY34, yielded 0.40±0.01 g ethanol/g of reducing sugars. The yeast isolate M. cibodasensisY34 appeared as promising ethanologen and embodies prospective potential for fermentative valorization of WMP-to-bioethanol.

Como em outros países agrícolas, grandes quantidades de cascas de melancia (WMP) são descartadas como resíduos no meio ambiente no Paquistão, mas a gestão adequada desses resíduos é a mais recente solução para salvar o meio ambiente da poluição. O trabalho enfatiza o papel das leveduras etanologênicas para utilizar açúcares significativos presentes no WMP para fermentação de bioetanol de baixo custo. A hidrólise de ácido clorídrico diluído de WMP foi realizada em condições otimizadas empregando RSM (metodologia de superfície de resposta) e seguindo o projeto de composto central (CCD). Este projeto experimental é baseado na otimização da etanologenesis envolvendo alguns parâmetros independentes importantes, como hidrolisado de WMP e razão de meio sintético (X1), temperatura de incubação (X2) e temperatura de incubação (X3) para rendimento máximo de etanol explorando o padrão (Saccharomyces cerevisiae K7) também como leveduras experimentais (Metchnikowia cibodasensis Y34). ABSTRACT

Endoscopic stent implantation is one of the main methods for the treatment of biliary and pancreatic diseases. At present, the commonly used biliary and pancreatic stents are mainly plastic and metal stents which are still have some deficiencies in clinical applications, and the emergence of the new type of biodegradable polymer materials is expected to achieve the purpose of treatment to overcome these shortcomings. It is a potential hope to break through the bottleneck of endoscopic treatment of choleopancreatic diseases. Previous animal experiments and human clinical studies have preliminarily shown its safety and effectiveness, which can effectively solve some problems of bile and pancreatic duct stenosis and so on. Biodegradable polymer stents have been widely studied, but their clinical application progress is slow and not yet popular, and it has gradually become a research hotspot in recent years . This article discusses the research status and development direction of biodegradable polymer stents in biliary and pancreatic diseases.

ABSTRACT

With the continuously increasing demands of plastic products in the current society, the challenge of disposing plastic waste is constantly increasing, leading to the urgent need of mitigating plastic pollution. As a consequence, much attention has been paid to biodegradable plastics due to their degradability in a bio-active environment under certain conditions. Biodegradable plastics herald vast development potentials and considerable market prospects. The degradation of numerous types of biodegradable plastics will be affected by many factors. A thorough understanding of degradation mechanisms as well as functional microbial strains and enzymes is the key to comprehensive utilization and efficient treatment and disposal of biodegradable plastics. The article summarized the types, properties, advantages and disadvantages, and main applications of common biodegradable plastics. The degradation mechanisms, functional microbial strains and enzymes, as well as the degradation degree and duration under different environmental conditions, were also summarized. This review may help better understand the degradation of biodegradable plastics wastes.

ABSTRACT

A multi-barrier antibiotics loaded biodegradable composite bone cement for resolving chronic osteo-myelitis has been studied to understand the physico-mechanical properties,drug loading/eluting effi-ciency,and different merits and demerits prior to clinical application.After successful induction of bone infection in 28 rabbits using methicillin-resistant Staphylococcus aureus(MRSA)strains,calcium sulfate/bioactive glass based composite cement was implanted in 12 defects to assess its performance over parenteral therapy with microscopic and radiological examination for 90 days.The composite cement revealed acceptable physico-mechanical properties and controlled drug elution kinetics.Furthermore,the antibiotics concentrations in bone up to 42 days were sufficient to kill MRSA without eliciting adverse drug reactions.The striking feature of platelets aggregation by composite cement could assist bone healing.The controlled degradation with simultaneous entrapment of composite cement within the osteoid tissues and complete repair of infected cortical defects(holes)in rabbit tibia at 6 weeks indicated the excellent anti-infective and osteoconductive properties of composite cement.Thus,the animal study demonstrated the superiority of composite over injectable antibiotic therapy based on infection reso-lution and bone regeneration.We thereby conclude that the composite cement can be effectively applied in the treatment of resistant cases of chronic osteomyelitis.

Objective To design a novel biodegradable intestinal anastomosis stent and explore the relationship between pressure distance and biomechanical properties of the anastomosis, so as to provide new ideas and methods for compression anastomosis of intestinal tissues. Methods A compression anastomosis stent was designed for reconstruction of intestinal tissues, and the finite element model of compression anastomosis of intestinal tissues was established to investigate the relationship between pressurized distance (2, 1.6, 1.2, 0.8 mm) and tissue stress of the stent. The anastomosis effect of intestinal tissue under different pressurized distances was analyzed by the test of tear-off force and burst pressure. Results Biomechanical properties of the intestinal anastomosis were the best when the pressurized distance was 1.2 mm, and its maximum tensile strength reached 0.77 MPa. The tear-off force and burst pressure of tissues were (25.80±1.82) N and (12.30±0.26) kPa, respectively. The optimal intestinal anastomosis was achieved when tissues were compressed to 60% of the original thickness. Conclusions The biodegradable intestinal stent designed in this study can successfully achieve compression anastomosis of intestinal tissues, which provides theoretical references for the development and application of novel biodegradable compression anastomosis devices.

Abstract In the hospital environment, postoperative pain is a common occurrence that impairs patient recovery and rehabilitation and lengthens hospitalization time. Racemic bupivacaine hydrochloride (CBV) and Novabupi® (NBV) (S (-) 75% R (+) 25% bupivacaine hydrochloride) are two examples of local anesthetics used in pain management, the latter being an alternative with less deleterious effects. In the present study, biodegradable implants were developed using Poly(L-lactide-co-glycolide) through a hot molding technique, evaluating their physicochemical properties and their in vitro drug release. Different proportions of drugs and polymer were tested, and the proportion of 25%:75% was the most stable for molding the implants. Thermal and spectrometric analyses were performed, and they revealed no unwanted chemical interactions between drugs and polymer. They also confirmed that heating and freeze-drying used for manufacturing did not interfere with stability. The in vitro release results revealed drugs sustained release, reaching 64% for NBV-PLGA and 52% for CBV-PLGA up to 30 days. The drug release mechanism was confirmed by microscopy, which involved pores formation and polymeric erosion, visualized in the first 72 h of the in vitro release test. These findings suggest that the developed implants are interesting alternatives to control postoperative pain efficiently.

Abstract Introduction Postoperative dysphonia is mostly caused by vocal fold scarring, and careful management of vocal fold surgery has been reported to reduce the risk of scar formation. However, depending on the vocal fold injury, treatment of postoperative dysphonia can be challenging. Objective The goal of the current study was to develop a novel prophylactic regenerative approach for the treatment of injured vocal folds after surgery, using biodegradable gelatin hydrogel microspheres as a drug delivery system for basic fibroblast growth factor. Methods Videoendoscopic laryngeal surgery was performed to create vocal fold injury in 14 rabbits. Immediately following this procedure, biodegradable gelatin hydrogel microspheres with basic fibroblast growth factor were injected in the vocal fold. Two weeks after injection, larynges were excised for evaluation of vocal fold histology and mucosal movement. Results The presence of poor vibratory function was confirmed in the injured vocal folds. Histology and digital image analysis demonstrated that the injured vocal folds injected with gelatin hydrogel microspheres with basic fibroblast growth factor showed less scar formation, compared to the injured vocal folds injected with gelatin hydrogel microspheres only, or those without any injection. Conclusion A prophylactic injection of basic fibroblast growth factor -containing biodegradable gelatin hydrogel microspheres demonstrates a regenerative potential for injured vocal folds in a rabbit model.


Resumo Introdução A disfonia pós-operatória é causada principalmente por cicatrizes nas pregas vocais. Tem sido relatado que o manejo cuidadoso da cirurgia das pregas vocais reduz o risco de formação de cicatriz. No entanto, a depender da lesão da prega vocal, o tratamento da disfonia pós-operatória pode ser desafiador. Objetivo Desenvolver uma nova abordagem regenerativa profilática para o tratamento de pregas vocais lesionadas após a cirurgia, com microesferas biodegradáveis de hidrogel de gelatina como sistema de administração de medicamentos para o Fator Básico de Crescimento de Fibroblastos (bFGF). Método A cirurgia laríngea videoendoscópica foi feita para criar lesão nas pregas vocais em 14 coelhos. Imediatamente após esse procedimento, microesferas biodegradáveis de hidrogel de gelatina com bFGF foram injetadas na prega vocal. Duas semanas após a injeção, as laringes foram excisadas para avaliação da histologia das pregas vocais e do movimento da mucosa. Resultados A presença de função vibratória deficiente foi confirmada nas pregas vocais lesionadas. A histologia e a análise de imagem digital demonstraram que as pregas vocais lesionadas injetadas com microesferas de hidrogel de gelatina com bFGF apresentaram menor formação de cicatriz, em comparação com as pregas vocais lesionadas injetadas apenas com microesferas de hidrogel de gelatina ou aquelas sem injeção. Conclusão Uma injeção profilática de microesferas biodegradáveis de hidrogel de gelatina com bFGF demonstra um potencial regenerativo para pregas vocais lesionadas em um modelo de coelho.

Resumen En términos generales, es bien conocida la cualidad que poseen algunos polímeros de cambiar sus propiedades físicas y químicas finales mediante la adición de nanopartículas a la matriz polimérica para producir un material compuesto (MC). Esta investigación está basada en la obtención de un MC a partir de ácido poliláctico (PLA) y nanotubos de carbono de pared múltiple (NTCPM), muy empleado en la industria del envasado y dispositivos biomédicos, con el fin de ampliar su perfil industrial. Se desarrollaron cuatro mezclas de PLA y NTCPM, y se empleó polietilenglicol (PEG) como plastificante. Se evaluaron sus propiedades morfológicas, térmicas, mecánicas, termo-mecánicas, espectroscópicas, ángulo de contacto y cristalográficas. Se observó que los MCs presentaron degradación térmica a temperaturas inferiores a la matriz sin NTCPM, así como un aumento en el módulo de flexión y tensión en algunas de las muestras. Así mismo, se observó que los NTCPM pueden aumentar la cristalinidad del material y que, en algunos casos, se incrementa su rigidez, actuando como un aditivo útil para aplicaciones de mayor esfuerzo mecánico que la matriz. Del efecto de agregar PEG en los MC, se determinó que los NTCPM no restringen la movilidad de las cadenas poliméricas y se da un efecto plastificante, lo que permite mayor movilidad de la zona amorfa de las cadenas de polímero, como indica la literatura consultada. Finalmente, se concluyó que a mayores contenidos de NTCPM, se generan mejores valores en el módulo de flexión, esfuerzo máximo de flexión, módulo de elongación, esfuerzo de carga máxima y esfuerzo de ruptura, entre otras propiedades evaluadas.


Abstract The quality of some polymers to change their final physical and chemical properties by adding nanoparticles to the polymer matrix to produce a composite material (MC) is well known. This research is based on obtaining a MC from polylactic acid (PLA) and multi-walled carbon nanotubes (CNTMW), widely used in the packaging industry and biomedical devices, in order to expand its industrial profile. Four mixtures of PLA and CNTMW were developed, and polyethylene glycol (PEG) was used as a plasticizer. Their morphological, thermal, mechanical, thermo-mechanical, spectroscopic, contact angle, and crystallographic properties were evaluated. It was observed that the composites showed thermal degradation at temperatures below the matrix without CNTMW, as well as an increase in the modulus of flexion and tension in some of the samples. Likewise, it was observed that the CNTMW can increase the crystallinity of the material and that, in some cases, its rigidity is increased, acting as a useful additive for applications of greater mechanical stress than the matrix. From the effect of adding PEG in the composites, the CNTMW do not restrict the mobility of the polymer chains and a plasticizing effect occurs, which allows greater mobility of the amorphous zone of the polymer chains. In general terms, it was concluded that at higher CNTMW contents, better values were generated in the flexural modulus, maximum flexural stress, elongation modulus, maximum load stress and rupture stress, among other evaluated properties.


Resumo Alguns polímeros têm a propriedade de alterar suas propriedades físicas e químicas finais, adicionando nanopartículas à matriz polimérica para produzir um composto. Esta pesquisa baseia-se na obtenção de composto partir de ácido polilático (PLA) e nanotubos de carbono de paredes múltiplas (MWCNT), amplamente utilizado na indústria de embalagens e dispositivos biomédicos, a fim de expandir seu perfil industrial. Foram desenvolvidas quatro misturas de PLA e MWCNT e o polietilenoglicol (PEG) foi usado como plastificante. Foram avaliadas suas propriedades morfológicas, térmicas, mecânicas, termo-mecânicas, espectroscópicas, ângulo de contato e cristalográficas. Observou-se que os compostos apresentaram degradação térmica em temperaturas abaixo da matriz sem MWCNT, além de aumento no módulo de flexão e tensão em algumas das amostras. Da mesma forma, observou-se que o MWCNT pode aumentar a cristalinidade do material e que, em alguns casos, sua rigidez é aumentada, atuando como um aditivo útil para aplicações de maior tensão mecânica que a matriz. A partir do efeito da adição de PEG nos compostos, determinou-se que o MWCNT não restringe a mobilidade das cadeias poliméricas e ocorre um efeito plastificante, que permite maior mobilidade da zona amorfa das cadeias poliméricas. Em termos gerais, concluiu-se que, com maiores teores de MWCNT, melhores valores foram gerados no módulo de flexão, tensão máxima de flexão, módulo de alongamento, tensão de carga máxima e tensão de ruptura, entre outras propriedades avaliadas.

ABSTRACT

Objective To study the effect of plaque eccentricity on stent performance and stress distributions of artery and plaque during stent implantation in stenotic vessels. Methods The stent and idealized stenotic vessels were constructed, and 4 different eccentricities (0%, 20%, 40%, 60%) were attributed to the plaque. Then the stent recoil, stent foreshortening, and stress distributions of artery and plaque when the stent was expanded to the target displacement were analyzed by the finite element method. Results Along with the increase of plaque eccentricity, both stent recoil and stent foreshortening gradually grew. At the same time, the stress of artery and plaque also showed an increasing tread, and high-stress areas gradually approached the narrow side. The maximum von Mises stress of the plaque was much greater than that of the artery. Conclusions Plaque eccentricity had a certain effect on performance of the stent and stress distributions of stenotic vessels. In stent design, the geometry of the plaque should be considered to improve clinical effect of the stent in interventional treatment.

Aims@#The primary aim of the present study is to evaluate the effect of rearing substrates on the nutritional content of black soldier fly larvae (BSFL) by incorporating Cupriavidus necator cells containing intracellular polyhydroxybutyrate (PHB) in BSFL diet to further increase the protein content and simultaneously to biologically extract the polymer by utilizing the digestive system of BSFL. The potential application of BSFL as a biological PHB extraction agent was determined.@*Methodology and results@#Two feeding strategies consists of a mixture of protein (P) to carbohydrate (C) with a ratio of P50:C50 food waste (control feeding) and feed with bacterial cells (modified feeding). A comparison on the proximate analysis between this research and two commercially available products were conducted. Feeding BSFL with P50:C50 food waste revealed the highest crude protein content of 81.3 ± 0.2%. Additional bacteria cells in the BSFL diet, however, showed a negligible decrease in crude protein content of 0.67% as compared to the control feeding. Howbeit, this results comparably higher in contrast to the commercial products, with increment of crude protein content by 12.1% and 40.8%, respectively.@*Conclusion, significance and impact of study@#Two desirable products were obtained from the feeding with cells: (1) high protein content of BSFL and (2) biologically extracted polymer. This is the first study to demonstrate the utilization of BSFL as a biological extraction agent to partially extract biopolymer and increase the protein content by feeding with cells.

Background: Menstrual cup is an alternative to sanitary pads, which has received attention in relatively small-scale studies in high income, and low- and middle-income countries, including among schoolgirls. Made of high-grade medical grade silicone (biodegradable) have the advantage of reuse, and can potentially last up to 10 years. Objective of this study was to assess and analyse the awareness regarding menstrual cup among the women of reproductive age group.Methods: Data collected from the selected eligible participants by an interviewer administered study proforma. It is a descriptive cross-sectional study over 1 month in women in reproductive age in A. J. Institute Mangalore, Karnataka, India.Results: A total 82% were aware about menstrual cup but only 2.6% have used it.Conclusions: One challenge of menstruation that is taken for granted in affluent countries is the simple question of how to manage or contain the menstrual flow and what happens to a girl or woman who is not able to do this successfully. Menstrual hygiene management is therefore an increasingly important (yet often unrecognized) issue that is heavily intertwined with girls’ education, empowerment, and social development.

Abstract Whey, a by-product of dairy industry, is a feedstock widely employed in the production of biodegradable films. However, these films present some limitations when considering the performance of synthetic polymers, especially biological transformation by decomposition. This work aimed to evaluate the effects of chitosan addition to whey-based films to improve films physical-chemical properties and resistance to microbial degradation. The results showed that there was an interaction effect between the chitosan concentration and the storage time for the physical-chemical properties of elongation at break and opacity. There was statistical difference among the formulations; however, for the moisture content and film thickness, there was no interaction effect between the formulation and the storage time. The films with 1.5 and 3.0 wt.% chitosan presented a yellowish hue, characteristic of the polysaccharide; this could also be detected by SEM analysis. The films presented an excellent biodegradability, being decomposed in about 8 days. Considering all chitosan contents tested had similar performances, the chitosan content of 0.15 wt.% was the one with the better cost-benefit relation.

ABSTRACT

Biodegradable stents (BDSs) are the milestone in percutaneous coronary intervention(PCI). Biodegradable polymeric stents have received widespread attention due to their good biocompatibility, moderate degradation rate and degradation products without toxicity or side effects. However, due to the defects in mechanical properties of polymer materials, the clinical application of polymeric BDS has been affected. In this paper, the BDS geometric configuration design was analyzed to improve the radial strength, flexibility and reduce the shrinkage rate of biodegradable polymeric stents. And from the aspects of numerical simulation, in vitro experiment and animal experiment, the configuration design and mechanical properties of biodegradable polymeric stents were introduced in detail in order to provide further references for the development of biodegradable polymeric stents.

Percutaneous intervention with occluder has become the preferred radical therapy for most of common congenital heart diseases,such as atrial septal defect (ASD),ventricular septal defect and patent ductus arteriosus.At present,most occluders for clinical use are made of nitinol,which will permanently exist in human body after implantation,and lead to the risk of severe complications such as chronic inflammation,cardiac perforation and high-grade atrioventricular block.Biodegradable occluder will be gradually replaced by body tissues after implantation.Investigators are exploring to develop biodegradable occluder with different biodegradable materials.In particular,a completely biodegradable atrial septum occlusion system (AbsnowTM) developed by the team of professor Zhang Zhiwei has been approved for clinical trial by China Food and Drug Administration (CFDA) in 2018,which is the first completely biodegradable atrial septal defect occluder entered the clinical trial phase in the world.The results of human trial test demonstrated the initial safety and effectiveness of this occluder.However,multi-center clinical trials are needed for evaluation and verification.Currently,selecting suitable materials for developing new biodegradable occluders,and the effectiveness and safety of occluders are the focuses and development trend of studies on biodegradable occluders in the treatment of congenital heart diseases.
